Gonna be honest, this whole piece is lacking a lot of ways.
Visually it's alright, you've got decent spritework, and the animation is fine stylistically, though the way it's integrated into the piece as a whole is awkward and jarring.
The humor is bland to put it nicely. There just isn't any punch to the jokes, the delivery is awful, and the pacing is rushed to the point where the entire routine seems all punchline and no buildup. Not to mention just about every single one of those punch-lines was dreadfully predictable thanks both to the give-away segment titles, and several tired set-ups and scenarios.
The spoken dialogue of the characters was absolutely unnecessary and served only to detract from the piece rather than enhance it in any real way. It felt tacked-on, half-hearted, and it did the worst thing possible anybody can do to even the best jokes, it EXPLAINED them. "Oh No, I'm not followin' you this time" "You got OWNED" "You know we're related right?" Each and every one of these lines made me groan from how PAINFULLY unnecessary they were. When it comes to comedy of this nature, less is certainly more in terms of dialogue. You have to let the action and the slapstick speak for itself, and if it can't, consider reworking the jokes.
Once again, the animation and the spritework shows promise, but you've really got to study up on comedic timing, delivery, and communication. Find the things that make you and others laugh and study them, find what makes them funny and try to implement it in your own work. As it stands, what you've got is a boring, sluggish mess of tired material with Donkey Kong slapped on in only the most superficial of ways.

Well, the jokes barely made me chuckle, the drawn bits weren't very good and Candy Kong is NOT related to Donkey Kong she's his girlfriend although in the Super Smash Bros Brawl trophy of Candy Kong it says that no one really knows if they are actually dating (I didn't lower your score for this but I feel like I must say it.)
The parodies were a little crap. The only semi funny thing was ironically the guy at the end saying "it's not funny". Read a book on humor. And I don't mean that negatively. It will help with your jokes.
I have no idea how this got front page because it's painfully unfunny.
