No idea who these people are, and after watching this crap, I definitely don't care.

Splendid humor!

It's all nicely drawn but it's more of a Animatic at this point then a animation. If you do post more like this all I really have to say is either add more frames or call it a Animatic.
Normally I wouldn't be this harsh in my ratings, but seeing all the praise that this video gets, makes me really ponder how much people actually do pay attention to the difference between this, and other works. This does look nice, sure, but for an animation, it lacks a huge amount of frames, sound quality is really shabby and in no way do I feel like these characters depict the actual characters accurately.
Let me go into some more detail here, to show what I mean.
First off, the frame rate. Counting the first 2 seconds, this has a total of 4 frames, the first 5 seconds, 7. For an animation to seem fluid to the eyes, you should at the very least do about 5 frames per second, to make the eye at least be ABLE to see this as an animation. This shouldn't be classified as an animation, but a picture reel at best. For simplicity, I'll continue to call it an animation, but it really does hurt me a little to use that word for this. It might really be worth it to screw down the quality of an individual frame, simply to up the amount of frames in the animation.
Then we have the audio. There are multiple issues I have with this, starting with the variation in clarity. Jinx's voice is often clear, well audible, while Syndra is honestly hard to properly hear if you don't up the volume a little. This is something I'm willing to overlook at times, but since it's getting 4-star ratings, I really want to press home every issue there is, that makes this less than a 4-star production to me.
Besides the clarity of the voice, none of the voices even remotely match those of the actual characters. Now, that can be written off as a form of "parody", but using an actual male voice for a female character just crosses that line for me.
Third issue with audio for me is how I do not feel like the actual voices oftentimes accurately match the images provided at that moment. Take 0:09, where Jinx looks more sad, worried, than "sarcastic" as she's apparently supposed to look like. Or 0:20, where Jinx doesn't have a trace of either sarcasm, worry or actually any emotion whatsoever on her face, and Syndra looks more befuzzled and confused than agitated, which her voice obviously is trying to convey. It's things like these that usually make me click away, but again, I just don't feel like this is worth 4 stars at all.
Now, as to actually properly parodying these characters, there needs to be some kind of link for me that tells me about what characters we are talking. That means I should be able to identify the characters we're speaking of, by their way of acting, their looks and their voices. I already made my point about the audio, but their way of acting also really doesn't come across as Jinx, Syndra or Lux whatsoever. Jinx is jittery, mischievous, always busy trying to blow things up. Syndra feels more omnipotent, godlike, looking down on others. Lux is happy, bouncy, to the edge of being annoying.
Now look through this video, and tell me, when you only know those traits about these people, would you be able to identify who is who? I sure as hell couldn't. It's that I recognize the skins they used for Jinx, which allowed me to deduce that the other character was Syndra, but besides that, there is practically no correlation between the actual in-game characters and this animation.
Now obviously this isn't a horrid thing. It looks okay, it has a decent punchline and it isn't too long. However, those things really do not outweigh the issues this animation has for me. It's good, but just not 4 star good. I'd rate this a 2.5, and really in the middle there. A 2 seems too blunt for me, but a 3 would indicate it's actually pretty good, which it also isn't. The only reason I voted a 2 on this thing is because I feel it really isn't being judged for what it is, and more as just a bunch of cute anime girls wearing skimpy, revealing clothes, or just having a huge rack. Again, nothing wrong with that, but it's not what this is supposed to be about when it's a 4 star animation.
Long story short, it's okay. Add more frames, work on the audio and actually read into the lore of these characters, and you're a lot further towards being a real good animation than you are right now.
