Those are not cupcakes but muffins! Derpy is very well known for her fondness of those.
Letters are just a joke - Derpy is often portrayed as mail delivery pony, but not very good at it, so the joke is that this kind of explains why.
Not only was that the cutest thing ever, it was very entertaining! It seemed though that Derpy would sometimes ignore the bubbles and swerve to the right :/

Derpy will never ignore bubbles unless she doesn't see them (and even a single pixel can make a difference between being in her line of sight and not) or you lead her into danger, which in turn means she will panic and run away from you for a moment. Remember, Derpy will literally be brighter if she sees them.
