As Lucky Number Slevin has already been mentioned (awesome film) I'll try and go with something that hasn't:
Slither is possibly the best comic horror film since Shawn of the Dead (Ok, so I'm not going too far back here). A fairly unknown cast shoved into a horror film about slugs that infest humans, this movie is full of laughs from start to finish. What makes it great is that throughout you aren't quite sure if it is serious- there is just enough horror and violence in there to make you think it is for real, but some things are just so surreal and hilarious that I for one just sat and giggled right the way through. It probably isn't for everyone- I guess there is a reason why nobody has mentioned it yet. Best watched when slightly drunk...
There are quite a few films mentioned in this thread that I haven't seen, and some that I haven't even heard of, so I'm going to do my best to rent a few of them out as well.

An excellent movie, shows that Steve Carrell can do more than just comedy.
The Departed was also quite good. You actually want to like Leonardo Dicaprio in it.

The film was all but sugarcoated. The first lines prove it : After Carrell's character just tried to kill himself, his sister arrives at the hospital to pick him up.
Sister : I'm so glad you're alive.
Carrell : At least one of us is...
From this point, I was totally hooked.
And yeah, the ending scene is maybe a bit sugarcoated, but as Eclectic pointed out, it's been a rough ride for the family, so it's just a good thing to end the movie like this.
One I wouldn't dare not nominate is Rocky 6 which will be as epic as the other.
I wouldn't dare to call it Rocky Balboa, it's Rocky 6. >:(
