It doesn't just have to be the most prolific, there's also style, victims selected, etc, the best serial killer to me is Dennis Rader, even though he didn't kill many people and is kinda stupid, he had a great style, went many years without being caught, and didn't rape.

Jack the Ripper. I'm using 'best' as the first one I think of when I hear serial killer.
He managed to kill off literally hundreds of soldiers, aliens, possibly scientists and security guards, then managed to hide for about twenty years, and then came back as a very prolific cop/soldier killer and, later, was responsible for assisting a revolution that isn't concluded to this day.
