At my current (side)job they're pretty cool about these things. If I say I have other plans already they'll just say "all right" and call someone else, regardless of how important those plans may be. Then again, they probably know that normally I'll be happy to cover someone's shift if I'm able to. If I would always say no they might start making a bigger deal out of it.
Anyway, in the situation you described, I guess what I would say depends a little on my recent history with covering other people's shifts. If I have to cover shifts regularly I'd stick with no. If it's a one-time thing I guess I'll go cancel or postpone the weekend, but only if he calls well beforehand.
By the way, if he literally says what you said I would hate his tone. Just ask me normally and don't give me that sarcastic bullcrap.
