Take your pick.

Adobe Flash and Toon Boom are the standbys for frame-by-frame animation, and Adobe After Effects joins the mix for tweened animations and effects. Those are the three to look into. For free software, you'll be looking at the free and open-source section, though I can't vouch for how good/bad any of those are.
