I'm creating an IOS app with CS5.5 and have been programming it as if the screen size will be fixed at 320*480px. What will I need to look out for in order for it to not look strange when played on an iPhone 5?

Your resolution is far too small. Apple have had retina displays in their devices since early 2010, and as such are soon dropping support for non-retina apps.
"Starting May 1, new apps and app updates submitted to the App Store must be built for iOS devices with Retina display and iPhone apps must also support the 4-inch display on iPhone 5." - Guidelines
