Well it depends on what you would like to do.
If you want to go site seeing, that is an easy question.
Battery park. financial district on maiden and wall st., Chinatown, little italy, prince street in soho, the east village, the flat iron building, (museum of sex is 2 blocks away), mid town has the empire state building and all the broadway shows and not to mention times square, then you hit central park which is very nice to relax in, after that you basically have your blue blood cafes and restaurants and theaters... unless your loaded there in no reason to go above 71st street. The best places in manhattan in mu opinion are the IMAX theatre and the Angelika Independent film theatre
FOOD?
i would stay in soho for food and wine, as well the meat packing district between 8th and 14th on the WAY east side, its the best in manhattan if you know the right places. STK house is most likely the best steak house i have ever been to as well as the awesome french italian cuban and thai restaurants below houston street on the west side.
Sport and activities.
Well i dont know much about that except for all the parks in manhattan like battery which has a nice bike trail, The park near NYU college that i cant remember the name of that has alot of musicians playing for money... my favorite act was a tabla and an acoustic guitar played like a slide dobro.. also chelsea piers has basically anything in sports you would want to do from vert skateboarding to batting cages.. And obviously central park, TONS of trails and Huge rock formations to climb all over and not to mention summer jam on the main stage in the park.. only problem is you need to bring brownies and not papers. HA!
GEtting drunk!?
For cheap stay in the east village. a bunch of punk and rickabilly bars are over there mixed with out of town yuppies.. yeah drinks are cheap and the crowds are fun there.
WINE?!?! my favorite place is a small place called "peasant" They have an awesome wine list with some of the best sicilian wine i have ever had and the absolute best rustic italian dishes that are almost as good as my grandmothers.
