I've had this problem occur before. You've probably been saving you flash or backing it up to another location (ie: a flash drive, external hardrive, or just another folder on your computer) and the location isn't plugged into your computer (flash drive etc.) or has been deleted (folder).

Try plugging in whatever you were using to save the .fla or .swf to and try again. If you've deleted the folder it was in, then look in the recycling bin. Otherwise you're screwed.
